Constitutive expression of cytochrome P450 s and glutathion S-transferase isozyme in tissue of several inbred mice

Abstract
The aim of current study was to identify constitutive expressions of CYP450 (1A2, 1B1 and 2E2) and GST (¥á and ¥ì) isozymes using RT PCR in the brain, lung, heart, liver, kidney and small intestine of seven inbred mice strains including A/J, BALB/c, C3H/He, C57BL/6, C57BL/10, CBA and DBA mice. In studying CYP gene mRNA expressions, the strongest CYP1A2 mRNA signal was detected in the liver of all inbred mice tested. However, it was hardly measurable the expression of CYP1 A2 mRNA in the brain, lung, heart and intestine. In CYP1B1 mRNA expression, relatively strong mRNA was detected in the kidney of all inbred mice tested, although the liver was negative for CYP1B1 mRNA expression. Constitutive CYP2E1 mRNA was also considerably expressed in all tissues tested except the brain. As compared with interstrains, C57BL/6 mouse brain showed noticeably enhanced expressions of CYP1B1 and CYP2E1 mRNA. In two GST gene mRNA expressions, the strongest GST ¥ámRNA expression was detectable in the liver. kidney and intestine of all inbred strains tested. Lower level of GST ¥áisoform was also existed in the heart and lung of normal inbred mice. A substantial interstrain difference in GST ¥ámRNA expression of the lung and heart was observed in our results. BALB/c and DBA mice showed very weak expression of GST ¥ámRNA in the lung and heart. In contrast. GST mRNA was comparatively well expressed in all tissues especially with strong expression in the liver, kidney and intestine of seven inbred strains. In conclusion, this study demonstrated that there were some tissue- and strain-ditJerences in the constitutive expressions of CYP and GST gene.
